    Targeted expression of human FSH receptor Asp567Gly mutant mRNA in testis of transgenic mice: role of the human FSH receptor promoter.

    AIM: To specifically express the Asp567Gly human follicle-stimulating hormone receptor (FSHR) under the control of its promoter to evaluate the phenotypic consequences in the presence of normal pituitary function. METHODS: We produced transgenic mice overexpressing the Asp567Gly human FSHR under the control of a 1.5kb 5'-flanking region fragment of its promoter. RESULTS: Mice were phenotypically normal and fertile. In males, mRNA could be detected in the testis and the brain, indicating that the 1.5kb promoter fragment drives expression not only in the gonads. The testis weight/body weight ratio and the testosterone levels in transgenic and non-transgenic littermates were similar. By in situ hybridisation we found that the transgenic FSHR was highly expressed in Sertoli cells, spermatocytes and round spermatids. However, a radioligand receptor assay failed to show a significant difference in total FSHR binding sites in testis homogenates of transgenic and wild type animals, suggesting that the transgenic FSHR is probably not translated into functional receptor protein. CONCLUSION: A 1.5kb 5'-region of the human FSHR drives mRNA expression of the transgene in the testis but leads to ectopic expression in germ cells and in the brain. No phenotypic consequences could be documented due to the lack of protein expression

    Interrelationships among predictors of automated vehicle acceptance: a structural equation modelling approach

    The study investigated the interrelationships between the UTAUT2 (Unified Theory of Acceptance and Use of Technology) constructs predicting intentions to use driverless automated shuttles. Survey data was gathered from individuals physically experiencing an automated shuttle in a mixed traffic environment on public roads in Trikala (Greece) as part of the CityMobil2 project. Structural equation modelling showed that intentions to use automated shuttles were most strongly driven by hedonic motivation, followed by performance expectancy and social influence. Performance expectancy mediated the relationship between effort expectancy and behavioural intention. This means that people's expectations of how difficult automated shuttles will be to use and their intentions to use them can be explained through their expectations around how these vehicles will perform. Technology savviness was a negative moderator of the relationship between social influence and performance expectancy and facilitating conditions, respectively. Tech-savvy individuals rely less on their social networks to nurture their beliefs that automated shuttles are useful and to have the necessary resources to use automated shuttles. Car use was a negative predictor of the intention to use automated shuttles. Future research should revisit the interrelationships between the UTAUT2 constructs and apply (quasi-) experimental studies to unravel the temporal interaction between constructs.Relevance to human factors/Relevance to ergonomics theoryThe study investigated the interrelationships between the UTAUT2 (Unified Theory of Acceptance and Use of Technology) constructs predicting intentions to use driverless automated shuttles.     The LIM-only protein FHL2 attenuates lung inflammation during bleomycin-induces fibrosis

    Fibrogenesis is usually initiated when regenerative processes have failed and/or chronic inflammation occurs. It is characterised by the activation of tissue fibroblasts and dysregulated synthesis of extracellular matrix proteins. FHL2 (four-and-a-half LIM domain protein 2) is a scaffolding protein that interacts with numerous cellular proteins, regulating signalling cascades and gene transcription. It is involved in tissue remodelling and tumour progression. Recent data suggest that FHL2 might support fibrogenesis by maintaining the transcriptional expression of alpha smooth muscle actin and the excessive synthesis and assembly of matrix proteins in activated fibroblasts. Here, we present evidence that FHL2 does not promote bleomycin-induced lung fibrosis, but rather suppresses this process by attenuating lung inflammation. Loss of FHL2 results in increased expression of the pro-inflammatory matrix protein tenascin C and downregulation of the macrophage activating C-type lectin receptor DC-SIGN. Consequently, FHL2 knockout mice developed a severe and long-lasting lung pathology following bleomycin administration due to enhanced expression of tenascin C and impaired activation of inflammation-resolving macrophages

    Drivers’ Intentions to Use Different Functionalities of Conditionally Automated Cars: A Survey Study of 18,631 Drivers from 17 Countries

    A number of studies have investigated the acceptance of conditionally automated cars (CACs). However, in the future, CACs will comprise of several separate Automated Driving Functions (ADFs), which will allow the vehicle to operate in different Operational Design Domains (ODDs). Driving in different environments places differing demands on drivers. Yet, little research has focused on drivers’ intention to use different functions, and how this may vary by their age, gender, country of residence, and previous experience with Advanced Driving Assistance Systems (ADAS). Data from an online survey of 18,631 car drivers from 17 countries (8 European) was used in this study to investigate intention to use an ADF in one of four different ODDs: Motorways, Traffic Jams, Urban Roads, and Parking. Intention to use was high across all ADFs, but significantly higher for Parking than all others. Overall, intention to use was highest amongst respondents who were younger (<39), male, and had previous experience with ADAS. However, these trends varied widely across countries, and for the different ADFs. Respondents from countries with the lowest Gross Domestic Product (GDP) and highest road death rates had the highest intention to use all ADFs, while the opposite was found for countries with high GDP and low road death rates. These results suggest that development and deployment strategies for CACs may need to be tailored to different markets, to ensure uptake and safe use
